How to enable the ports on my motherboard

Hello,
I have 3 PCI ports on my motherboard (2 PCI x1 and 1 PCI x16) but only one PCI x1 is active. The other two are greyed out in the BIOS and I haven't found any way to activate them.
Is it possible?

There is nothing special to do for the PCI-Express 16X ports on a motherboard to work. If the one on your motherboard doesn't work, it may be that your port is faulty. What motherboard do you have (brand and model)?

Thank you for getting back to me.
In fact, if I plug another card into slot 2 (slot 1 is occupied by a PCI card with 4 SATA ports), I can no longer boot from the drives connected to the card in slot 1 and I no longer have access to the BIOS boot menu.

The card I wanted to install in slot 2 is a USB 3 port card.

The two SATA ports on my motherboard have become disconnected and I only have those on the card in slot 1 to operate the computer.

The PCI ports on the motherboard do not seem to be defective indeed.

Thank you for your help, which at least allowed me to see things a bit clearer.

It seems that your card is not recognized or there is a conflict between the two USB and SATA cards.

I assume you haven't been able to install drivers after installing the cards, right?

I don't currently have a solution to resolve this issue. However, if I can suggest something, it's to do a clean install of the drivers for your SATA card (I think the model name would be useful to share), then adjust the Boot settings in the BIOS if needed, and then insert the USB card and follow the same process.

Hello,
the USB 3 card can also be defective.
There’s nothing to activate in the BIOS; if it’s grayed out, it means it doesn’t detect anything plugged in.

When you talk about slot 1 and 2, are you referring to the x1 slots?

If you plug your SATA card alone into slot 2, does it work? (or at least can you access the BIOS?)

If you plug the USB card alone into slot 1 or 2, can you access the BIOS?
