Hello, when you block someone for an hour and then unblock them afterwards, do you receive the messages they sent during the hour you had them blocked after unblocking them?

Hello, the answer is NO, all the messages that your contact sent you during the block are directly deleted from the iPhone and there is no way to recover them, even if you unblock the contact.

Is the blocked contact informed of the rejection of their messages?

The contact is not normally informed.
However, on certain phones, when you unblock the person, you receive all the messages that were sent.
